• 白细胞介素--22促进类风湿关节炎成纤维化膜细胞的增殖

    Abstract: Objective To clarify the mechanism by which interleukin-22 (IL-22) promotes the proliferation of fibroblast-like synoviocytes (FLS) from patients with rheumatoid arthritis (RA). Methods FLS were isolated from the synovial tissues of patients with RA and identified by immunohistochemistry for vimentin/CD68. The cells were subcultured and incubated with different concentrations of IL-22 for 24, 48, or 72 h, and their proliferation was examined using MTT assay. After treatment of the cells with IL-22 and AG490, alone or in combination, the expressions of the total and phosphorylated proteins of STAT3, ERK1/2 and P38 were detected with Western blotting. Results IL-22 significantly increased the proliferation of FLS in a dose-dependent manner (P<0.05). The total protein of STAT3 in the cells showed no significant changes with extended time of IL-22 treatment (P=0.68), but the expression of phosphorylated STAT3 protein increased significantly (P<0.001). The total and phosphorylated proteins of ERK1/2 and P38 underwent no significant changes after IL-22 treatment (P>0.05). A combined treatment with 50 ng/mL IL-22 and 100 μmol/L AG490 resulted in a significant decrease in the proliferation of FLS as compared with IL-22 treatment alone (P<0.01). Conclusion IL-22 can dose-dependently promote the proliferation of FLS from patients with RA by inducing phosphorylation of STAT3 protein but not through ERK1/2 or P38 signal pathway.

    Abstract: Objective To observe the effect of rhynchophylline on methamphetamine-dependent zebrafish and explore the possible mechanism. Methods Zebrafish were divided into control group, amphetamine group, low- (50 mg/kg) and high (100 mg/kg)-dose rhynchophylline groups, and ketamine (150 mg/kg) group. Conditioned place preference (CPP) was induced in zebrafish with methamphetamine, and the staying time in the drug box and the tracking map of the zebrafish were observed with Noldus Ethovision XT system. The protein expressions of TH, NR2B and GLUR2 in the brain of zebrafish with CPP were detected with Western blotting. Results Compared with the control group, zebrafish in methamphetamine group showed significant variations in the staying time and swimming distance in the drug box after conditioning (P<0.05) with obvious alterations of NR2B, TH and GLUR2 expressions in the brain (P<0.05). Treatment of methamphetamine-dependent zebrafish with high-dose rhynchophylline significantly reduced the variations in the staying time and swimming distance in the drug box (P<0.05) and in the expressions of NR2B, TH and GLUR2 in the brain (P<0.05). Conclusion Rhynchophylline can inhibit methamphetamine dependence in zebrafish, the mechanism of which may involve the expressions of TH, NR2B and GLUR2 proteins in the brain.

  • 三黄茵赤方含药血清对LO2细胞DNA氧化损伤的影响

    Abstract: Objective To study the protective effect of Sanhuangyinchi Fang drug serum (SF) against hydrogen peroxide-mediated DNA oxidative damage in LO2 cells. Methods The LO2 cells were randomly divided into the control group , H2O2 group, SF groups (5%, 10%, and 15%) and vitE group. The morphological features of the treated LO2 cells were observed under inverted microscope. The viability of the treated cells was assessed with CCK-8 method, and the activity of SOD, CAT and GSH-PX were detected biochemically. Reactive oxygen species (ROS) levels, the content of 8-OHdG, and DNA damage of the cells were evaluated by flow cytometry, ELISA, and Comet assay, respectively. Results Compared with H2O2 group, the cells in SF groups (10% and 15%) and vitE group showed higher cell survival rate (P<0.05) and higher SOD, CAT, GSH-PX (P<0.05) and ROS scavenging activities (P<0.01) with markedly decreases the content of 8-OHdG (P<0.01) and reduced tailing ratio, tail length, tail moment and Olive tail moment (P<0.05). Conclusion SF drug serum, especially at the concentration of 15%, can protect LO2 cells from H2O2 -mediated DNAoxidative damage.
