摘要: In this paper, low-energy scattering of the (D∗D¯∗)± meson system is studied within L\"uscher's finite-size formalism using Nf=2 twisted mass gauge field configurations. With three different pion mass values, the s-wave threshold scattering parameters, namely the scattering length a0 and the effective range r0, are extracted in JP=1+channel. Our results indicate that, in this particular channel, the interaction between the two vector charmed mesons is weakly repulsive in nature hence do not support the possibility of a shallow bound state for the two mesons, at least for the pion mass values being studied. This study provides some useful information on the nature of the newly discovered resonance-like structure Zc(4025) observed in various experiments.

摘要: Long noncoding RNAs (IncRNAs), which have emerged in recent years as a new and crucial layer of gene regulators, regulate various biological processes such as carcinogenesis and metastasis. HOTAIR (Hox transcript antisense intergenic RNA), a lncRNA overexpressed in most human cancers, has been shown to be an oncogenic lncRNA. Here, we explored the role of HOTAIR in HeLa cells and searched for proteins regulated by HOTAIR. To understand the mechanism of action of HOTAIR from a systems perspective, we employed a quantitative proteomic strategy to systematically identify potential targets of HOTAIR. The expression of 170 proteins was significantly dys-regulated after inhibition of HOTAIR, implying that they could be potential targets of HOTAIR. Analysis of this data at the systems level revealed major changes in proteins involved in diverse cellular components, including the cytoskeleton and the respiratory chain. Further functional studies on vimentin (VIM), a key protein involved in the cytoskeleton, revealed that HOTAIR exerts its effects on migration and invasion of HeLa cells, at least in part, through the regulation of VIM expression. Inhibition of HOTAIR leads to mitochondrial dysfunction and ultrastructural alterations, suggesting a novel role of HOTAIR in maintaining mitochondrial function in cancer cells. Our results provide novel insights into the mechanisms underlying the function of HOTAIR in cancer cells. We expect that the methods used in this study will become an integral part of functional studies of IncRNAs.

摘要: Mycobacterium tuberculosis (Mtb), the causative agent of human tuberculosis, remains one of the most prevalent human pathogens and a major cause of mortality worldwide. Metabolic network is a central mediator and defining feature of the pathogenicity of Mtb. Increasing evidence suggests that lysine succinylation dynamically regulates enzymes in carbon metabolism in both bacteria and human cells; however, its extent and function in Mtb remain unexplored. Here, we performed a global succinylome analysis of the virulent Mtb strain H37Rv by using high accuracy nano-LC-MS/MS in combination with the enrichment of succinylated peptides from digested cell lysates and subsequent peptide identification. In total, 1545 lysine succinylation sites on 626 proteins were identified in this pathogen. The identified succinylated proteins are involved in various biological processes and a large proportion of the succinylation sites are present on proteins in the central metabolism pathway. Site-specific mutations showed that succinylation is a negative regulatory modification on the enzymatic activity of acetylCoA synthetase. Molecular dynamics simulations demonstrated that succinylation affects the conformational stability of acetyl-CoA synthetase, which is critical for its enzymatic activity. Further functional studies showed that CobB, a sirtuin-like deacetylase in Mtb, functions as a desuccinylase of acetyl-CoA synthetase in in vitro assays. Together, our findings reveal widespread roles for lysine succinylation in regulating metabolism and diverse processes in Mtb. Our data provide a rich resource for functional analyses of lysine succinylation and facilitate the dissection of metabolic networks in this life-threatening pathogen.

摘要: The recent emergence of liquid crystals of atomically thin two- dimensional (2D) materials not only has allowed us to explore novel phenomena of macroscopically aligned 2D nanomaterials but also has provided a route toward their controlled assembly into three-dimensional functional macrostructures. Using flow- induced mechanical alignment, we prepared flakes of graphene oxide (GO) in different orientational orders and demonstrated that GO liquid crystals (LC) can be used as rewritable media for reflec- tive displays without polarizing optics. With a wire or stick as a pen, we can make the surface of GO LC reflective and bright, and we can then manually draw lines, curves, and any patterns with dark appearance. The contrast between bright and dark features is due to anisotropic optical responses of ordered GO flakes. Since optical anisotropy is an intrinsic property of 2D structures, our observations and demonstration represent one of many potential applications of macroscopically aligned 2D nanomaterials.
