  • 添加剂对大孔吸附树脂固定化脂肪酶的影响

    Abstract:利用大孔吸附树脂DA-201为载体对海洋脂肪酶固定化,并探寻添加剂对固定化过程的影响。分别以NH4Cl、甘露糖和甘氨酸为添加剂,采用单因素和正交实验相结合的方法优化条件。结果显示,以NH4Cl为添加剂的最优条件:柠檬酸-柠檬酸钠缓冲液pH 6.0,固定化温度30℃,载体投放量0.5g,NH4Cl浓度为25mmol/L,固定化时间3.0h,酶活力达到115.27U/g;比不含有添加剂的固定化酶固定化效率提高47.42%。以甘露糖为添加剂最有条件:磷酸二氢钾-氢氧化钠缓冲液pH7.0,固定化温度35℃,载体投放量0.5g,甘露糖浓度10mmol/L,固定化时间4.5h;酶活力达到122.75U/g,比不含有添加剂的固定化酶固定化效率提高6.50%。以甘氨酸为添加剂的最优条件:磷酸二氢钾-氢氧化钠缓冲液pH7.0,固定化温度20℃,载体投放量0.5g,甘氨酸浓度为25mmol/L,固定化时间7.5h;酶活力达到141.69U/g,比不含有添加剂的固定化酶固定化效率提高26.12%。采用不同添加剂对大孔吸附树脂DA-201的吸附固定化过程有较大影响,可以极大的提高吸附效率;同时发现缓冲液类型、pH、温度、添加剂浓度和固定化时间等对DA-201树脂吸附脂肪酶有很大影响,对后续吸附固定化工业酶研究有较好的参考价值。

    Abstract: The Plagiogyria is isolated in the classification system, an so far no reports about the systematic and comparative study of its anatomical structure have been published. The anatomical comparative study of the petiole and the pinnule cross-sections of 9 species of Plagiogyria was performed using light microscopy for the first time. The epidermis, the basic tissue and the middle column of the middle petiole transection and the epidermis, mesophyll and main vein of the vegetative leaf pinnule of 9 species of Plagiogyria are similar, for example, the epidermis of the transverse section of the middle petiole is not covered with hair or squamas, the epidermis is round in shape with thick-walled tissue, and the central cylinder is a amphicribral vascular bundle; the stomatas of trophyll pinnule transection are only distributed in the lower epidermis, the shape of epidermal cells is flat, and the type of the main vein belongs to the peripheral vascular bundle, which supports for the viewpoint that Plagiogyria is a natural taxonomy. In the evolution of the system, the Plagiogyria has a certain relationship with the Alsophila spinulosa and both of them have similar characteristics and also show certain differences. However, the petiole central transverse section of 9 species of Plagiogyria are trapezoidal, oval or triangular. The shape of the vascular bundle including "U" shape, "V" shape or triangle. The number of vascular bundles in the central transverse section of the petiole is one or three. The arrangement of xylem in vascular bundles including "Eight" shape, "U" shape or " 一" shape; The underside protuberances of the pinnule main veins are curved, triangular or trapezoidal. The xylem of the petiole has two forms: typical hippocampus and atypical hippocampus. The typical hippocampus is characterized by obvious hooks state at both ends of the xylem. The atypical hippocampus is characterized by no obvious hook state at the two ends of xylem or one of the xylems has no hook at one end. These morphological anatomical features are stable and cluster specific, providing a new reference for taxonomy and systematic studies of Plagiogyria. Finally, a key to the Plagiogyria species is given based on the anatomical characteristics of the transverse section of the petiole and the pinnule.

  • 皮内注射亚甲蓝治疗老年胸段中重度带状疱疹

    Abstract: Objective To evaluate the clinical efficacy of intradermal injection of methylene blue for treatment of moderate to severe acute thoracic herpes zoster and prevention of postherpetica neuralgia in elderly patients. Methods Sixty-four elderly patients with herpes zoster were randomized to receive a 10-day course of intradermalinjectionof methylene blue and lidocaine plus oral valaciclovir (group A, 32 cases) and intradermalinjectionof lidocaine plus oral valaciclovir (group B). Herpes evaluation index, pain rating index, incidence of postherpetic neuralgia, and comprehensive therapeutic effect were compared between the two groups at 11, 30 and 60 days after the treatment. Results The baseline characteristics were comparable between the two groups (all P>0.05). Compared with that in group B, the time for no new blister formation, blister incrustation and decrustation, and pain relief was significantly shortened in group A (P<0.05) with also obviously lower pain intensity after the treatment. The incidence of postherpetic neuralgia was significantly lower in group Athan in group B at 30 days (P<0.05), but not at 60 and 90 days after the treatment. The total clinical response rate was 93.8% in groupA, much higher than that in group B (62.5%, P<0.05). Conclusion Intradermalinjectionof methylene blue can effectively shorten the disease course,reducethepainintensityandpreventthedevelopmentofpostherpeticneuralgiainelderlypatientswithherpeszoster.

    Abstract:以海藻酸钠、羧甲基纤维素钠(CMC)为载体,分别以乙二醇缩水甘油醚(EGDE)和戊二醛为交联剂,采用包埋交联法对脂肪酶进行固定化,结果显示EGDE的交联效果要优于戊二醛,添加EGDE的固定化酶酶活最好。得到制备固定化酶的最优方案为海藻酸钠2.5%,CMC 浓度1.5%,给酶量800 U/mL复配载体,氯化钙5%,以0.02%的EGDE交联固定30 min,由此制备得到酶活约为380 U/g的固定化酶。固定化酶的最适反应pH为8.5,比游离酶增大0.5个单位;最适反应温度是45℃,比游离酶提高5℃;耐热性能变好,且重复使用7次后仍能保持60%左右的相对酶酶活。
