    Abstract:随着科技的进步,媒介传播技术不断得到发展,我们已经进入数字媒体时代。与此同时,报纸、广播、电视等传统媒体面临生存和发展的考验。手机、互联网等数字媒体的出现使得传统媒体受众数量减少、广告收入下滑。这一现象受到学界和业界的广泛关注,本文在具体实例的基础上,从内容和渠道两个方面,探析在数字媒体时代背景下电视媒体该如何发展。

    Abstract: Updating outdated memories with newly acquired information is an important ability. To ensure less interference from outdated memories, one either trims old memory traces or differentiates between novel and old memories. Although the age-related decline in episodic memory has been well-established, aging effect on episodic memory updating and its underlying cognitive mechanisms are less understood. Memory differentiation and memory interference may act as two different mechanisms that underlie episodic memory updating. To bridge this research gap, we investigated the different updating memory patterns in young and older adults. Furthermore, we intended to clarify the updating differences in item and source memory, which are the two essential elements embedded in episodic memory. By implementing an adapted AB-AC memory updating paradigm, we posited that young adults would comparably attain A-B and A-C memory. In contrast, older adults would experience C memory intrusions in the A-B memory test, indicating memory interference. We examined episodic memory updating for a sample of 30 young and 30 older adults randomly selected from the university and neighboring community. On Day 1, participants learned 36 naturalistic A-B pairings and completed the encoding test immediately after learning. On Day 2, participants first reviewed the A-B pairs before being introduced to 36 novel A-C pairings (where C’s items or sources were different from those in B), following which they completed the Day 2 encoding test. Finally, all the participants returned to the laboratory and completed the A-B and A-C tests on Days 3 and 5. To elucidate the updating memory patterns between the two age groups, we classified their responses into target, competitor, and lure categories to test the group-level differences in memory updating. We ran a 2 (age: young adults and older adults) × 2 (association: A-B and A-C) × 3 (category: target, competitor, and lure) × 2 (time: Days 3 and 5) repeated analysis of variance to examine the different memory updating pattern between the two age groups. Expectedly, there were significant age×category and age×association×category interaction effects in both item and source memory tasks. More precisely, young adults performed comparatively well in A-B and A-C memory tests and showed no significant memory intrusions and distortions. In comparison, older adults performed better in the A-C memory test than in the A-B memory test and showed B intrusions in the A-C memory test but no C intrusion in the A-B test. We found that young adults could strengthen their novel memories without inhibiting outdated ones and simultaneously maintained A-B and A-C memory traces. However, older adults updated their memories by overwriting their previously-stored A-B memory traces. The findings illuminate episodic memory updating and its underlying cognitive mechanisms, whereby young adults update memories via differentiating old with new memory traces. However, older adults inhibit previous memory traces when confronted with two pieces of competing information. Notably, separate mechanisms apply to item and source memories and show a long-term effect.

  • 酵母多糖对哺乳犊牛胃肠道发育及消化酶活性的影响

    Abstract:本试验旨在研究饲粮中添加不同剂量的酵母多糖对哺乳犊牛胃肠道发育及消化酶活性的影响。选择初生重相近的健康中国荷斯坦犊牛56头,随机分为4组,每组14头。Ⅰ组(对照组)饲喂基础饲粮,Ⅱ、Ⅲ、Ⅳ组在饲喂基础饲粮中分别添加1、2、3 g/(头·d)酵母多糖。试验期60 d。结果表明:1)Ⅱ、Ⅲ、Ⅳ组犊牛瘤胃淀粉酶、胃蛋白酶、脂肪酶和羧甲基纤维素酶活性高于Ⅰ组,Ⅲ组显著高于Ⅰ组(P<0.05);Ⅲ组犊牛十二指肠淀粉酶、胰蛋白酶、脂肪酶、羧甲基纤维素酶活性显著高于Ⅰ组(P<0.05);Ⅲ组犊牛空肠淀粉酶(P<0.05)、胰蛋白酶(P<0.01)活性显著或极显著高于Ⅰ组,Ⅱ、Ⅲ、Ⅳ组空肠脂肪酶、羧甲基纤维素酶活性显著高于Ⅰ组(P<0.05)。2)Ⅲ组犊牛瘤胃乳头长度、宽度和黏膜厚度显著高于Ⅰ组(P<0.05);十二指肠、空肠中段绒毛高度显著高于Ⅰ组(P<0.05),隐窝深度显著低于Ⅰ组(P<0.05),绒毛高度/隐窝深度(V/C)显著高于其他各组(P<0.05)。由此可见,添加酵母多糖能促进犊牛胃肠道形态发育,提高胃肠道消化酶活性。在本试验条件下,哺乳犊牛饲粮中酵母多糖的适宜添加量为2 g/(头·d)。

  • 饲粮铜水平对奶牛生产性能、养分表观消化率及血清生化指标的影响

    Abstract:本试验旨在研究饲粮铜水平对奶牛生产性能、养分表观消化率及血清生化指标的影响。选取60头产奶量、泌乳天数及胎次相近的中国荷斯坦奶牛,按随机分配原则分为4组,各组饲粮中铜添加量分别为0(对照,Ⅰ组)、10(Ⅱ组)、15(Ⅲ组)和20 mg/kg(Ⅳ组),饲粮铜水平分别为8.09、18.09、23.09和28.09 mg/kg DM,每组15头,试验期共93 d。结果表明:1)与Ⅰ组相比,Ⅱ、Ⅲ与Ⅳ组的产奶量分别提高了4.91%、6.27%及4.78%(P<0.05),乳中体细胞数量极显著降低(P<0.01)。2)与Ⅰ组相比,Ⅱ和Ⅲ组的中性洗涤纤维(NDF)表观消化率分别提高了6.21%和7.60%(P<0.05),酸性洗涤纤维(ADF)表观消化率分别提高了6.78%和8.22%(P<0.05)。3)与Ⅰ组相比,Ⅱ、Ⅲ和Ⅳ组血清铜、铜蓝蛋白含量及超氧化物歧化酶(SOD)、铜-锌超氧化物歧化酶(Cu-Zn SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性极显著提高(P<0.01),丙二醛(MDA)含量分别降低了50.00%、60.23%和47.08%(P<0.01),免疫球蛋白G(IgG)含量分别提高了49.56%、165.56%和90.22%(P<0.01)。4)铜代谢指标分析说明,与Ⅰ组相比,饲粮中补充铜能极显著提高粪铜、尿铜含量(即粪和尿中铜的排出量)和铜沉积量(P<0.01),但饲粮铜水平对铜表观消化率没有显著影响(P˃0.05)。在本试验条件下,饲粮中补充铜可提高奶牛的生产性能,促进机体对养分的利用,提高机体免疫力和抗氧化能力,但粪铜随着饲粮铜水平的增加而增加。综合考虑,在本试验条件下,奶牛饲粮适宜铜水平为18.09~23.09 mg/kg DM(铜采食量为403.59~514.21 mg/d)。

  • 饲粮全棉籽比例对荷斯坦公牛育肥性能、血清生化指标和养分表观消化率的影响

    Abstract:本试验旨在研究饲粮全棉籽比例的对荷斯坦公牛育肥性能、血清生化指标和养分表观消化率的影响。选取44头体重[(286±52) kg]相近、健康无疾病的荷斯坦公牛,随机分为4组,每组11头,Ⅰ、Ⅱ、Ⅲ和Ⅳ组分别饲喂含有0、5%、10%和15%全棉籽的饲粮,各组能量和粗蛋白质水平基本相同。预试期为7 d,试验期为90 d。结果表明:1)Ⅳ组平均日增重最高,比Ⅰ组提高了9.80%(P<0.05);Ⅳ组的干物质采食量最高,料重比最低,但各组间差异均不显著(P>0.05)。2)Ⅲ和Ⅳ组的血清胰岛素样生长因子1含量分别比Ⅰ组高4.79%和6.76%(P<0.05);血清中谷丙转氨酶、谷草转氨酶活性及游离脂肪酸和生长激素含量随全棉籽比例提高而有所上升,但组间并没有显著差异(P>0.05)。3)随着全棉籽比例的增加,中性洗涤纤维、酸性洗涤纤维、钙的表观消化率均有所提高,但各组间无显著

  • 家蝇抗菌肽AMPs17蛋白原核表达条件的优化及其抗真菌活性检测

    Abstract:目的:优化AMPs17重组蛋白的原核表达条件,分析重组蛋白的抗真菌活性。方法:比较不同的诱导温度(25 ° C, 28 ° C, 30 ° C, 32 ° C, 34 ° C)、异丙基硫代-β-D半乳糖苷(IPTG)诱导浓度(0.025、 0.05、0.1、0.3、0.5、0.8、1.0mmol·L-1)和诱导时间(12h、15h、18h、21h、24h)对AMPs17重组蛋白表达量的影响,筛选AMPs17重组蛋白的最佳表达条件;采用镍离子金属螯合剂亲和层析柱对重组蛋白进行纯化,SDS-PAGE电泳和ImageJ图像分析系统对表达结果进行分析,Western bolt对AMPs17重组蛋白鉴定,高效液相色谱分析重组蛋白的纯度,微量液体稀释法及菌落计数法检测其抗真菌活性。结果:在诱导温度为32℃、IPTG浓度为0.05mmol·L-1的条件下诱导培养15h,AMPs17重组蛋白的表达量最高且最为稳定;HPLC色谱仪分析显示AMPs17重组蛋白纯度可达到90%以上;优化后的AMPs17重组蛋白能有效抑制白色念珠菌的生长。结论:本研究优化了家蝇抗菌肽AMPs17的诱导表达条件,获得了高表达、稳定且具有抗真菌活性的蛋白,为后续抗菌机制及应用研究提供一定的实验基础。

  • 过表达DLL3促进人胃癌细胞的增殖

    Abstract: Objective To construct a eukaryotic expression plasmid carrying human full-length Notch ligand Delta-like 3 (DLL3) gene and study the effect of DLL3 knockdown and overexpression on the proliferation of gastric cancer cells in vitro. Methods Human full-length DLL3 gene was amplified by PCR and cloned into the eukaryotic expression vector pCMV-Tag4. After verification by restriction enzymes and sequencing, the recombinant DLL3/pCMV-Tag4 vector was transiently transfected into HEK293T cells, in which the expressions of human DLL3 mRNA and protein were detected using real-time quantitative PCR and Western blotting, respectively. The expression of DLL3 in normal gastric epithelial cells and gastric cancer cell lines was detected by qRT-PCR and Western blotting. DLL3/pCMV-Tag4 was transfected into 3 gastric cancer cell lines, and their proliferation was assessed with MTT assay. Human gastric cancer cells MGC803 and MKN45 were also transfected with a specific human DLL3-siRNA to assess the effect of DLL3 down-expression on the cell proliferation. Results The recombinant eukaryotic expression vector DLL3/pCMV-Tag4 was successfully constructed and human full-length DLL3 was expressed in HEK293T cells. MTT assay showed that DLL3 over-expression obviously promoted the proliferation and down-regulation of DLL3 inhibited the proliferation of the gastric cancer cells. Conclusion DLL3 overexpression can promote the proliferation of gastric cancer cells in vitro, and down-regulation of DLL3 inhibits the proliferation of gastrc cancer cells,which provides a novel strategy for targeted thrapy of gastric cancer.

  • 串列式叶栅扩压器非定常流动研究

    Abstract:采用数值模拟方法,对串列式叶栅扩压器中的非定常流动进行了研究。结果表明,带串列式叶栅扩压器的离心压气机内部流动是以强非定常的三维黏性主流和缝隙流动为主要特征,串列式叶栅扩压器内部流动存在明显的非定常性,压力脉动的传播在前排叶栅通道内衰减很快。扩压器前排叶片通道内压力脉动的主频为叶轮叶片通过频率。后排叶片前缘附近压力脉动的主频变为0.5倍叶频。串列式叶栅扩压器进口的压力脉动主要受到叶轮的激励作用;串列叶栅间隙处,随主流输运而来的叶轮尾迹与缝隙流、前排叶片尾迹相互干扰与叠加,成为后排叶片前缘压力脉动新的激励。相比于近叶根、叶尖处,前排叶片前缘中间叶高处的压力脉动受到叶轮主流和分流叶片尾迹的激励作用更强烈。

  • 围产前期饲粮粗蛋白质水平对荷斯坦经产奶牛产后生产性能和血液指标的影响

    Abstract:本试验旨在研究围产前期饲粮粗蛋白质水平对产后奶牛生产性能和血液生化指标影响。选取预产期、胎次、体重相近,上一个泌乳期时长、产奶量相近的21头经产荷斯坦奶牛,随机分为3组,每组7头。在围产前期(产前21 d内)饲喂粗蛋白质水平分别为12.12%(A组)、13.07%(B组)、14.02%(C组),产奶净能均为5.50 MJ/kg的3种不同饲粮,研究其对奶牛产后1~30 d的生产性能和血液指标的影响。结果表明:1)产后1~30 d,B组的干物质采食量分别比A、C组高5.41%、5.85%(P0.05)。产后2~10 d,B组的乳糖率比C组高4.31%(P<0.05)。围产前期B组的粗蛋白质表观消化率比A组高12.56%(P<0.05)。2)产后10、20、30 d,B组血清葡萄糖(GLU)含量分别比C组高31.41%、29.47%、21.38%(P<0.05)。分娩当天,B组血清总蛋白(TP)含量比C组高12.98%(P<0.05)。产后30 d,B组血清白蛋白(ALB)、瘦素(LP)含量分别高于C组14.69%、5.97%(P<0.01)。产后20 d,B组血清甘油三酯(TG)含量均比A、C组低3.57%(P<0.05),血清胰岛素(Ins)含量分别比A、C组低7.26%、12.47%(P<0.01)。产后30 d,B组血清胰岛素样生长因子Ⅰ(IGF-Ⅰ)含量比C组低4.94%(P<0.01)。综上,饲粮粗蛋白质水平13.07%为围产前期适宜饲粮粗蛋白质水平。
