  • Analysis and Identification of Hub Genes in Hepatocellular Carcinoma Based on Weighted Gene Co-expression Network and Cancer Genome Atlas Clinical Data

    Abstract: Background Hepatocellular carcinoma(HCC) is the third leading cause of common cancer-related mortality globally,accounting for approximately 90% of all primary liver cancer cases. Its recurrence and mortality rates are high,with the underlying molecular mechanisms remaining unclear. Objective To explore potential molecular mechanisms of HCC and explore novel biomarkers. Methods Gene expression profile GSE62232 was retrieved from the GEO database,RNA-seq expression data and clinical information were retrieved from TCGA database,differential gene expression analysis was conducted between normal liver tissue and HCC tissue. Enrichment analysis on the differentially expressed genes was performed. Based on the gene expression data profiles of HCC and GSE62232 in TCGA,a co-expression network was established using the WGCNA R package,and weighted gene co-expression network analysis(WGCNA) was performed to select clinically significant modules and screen candidate Hub genes;the candidate Hub genes were further analyzed for significant differential expression in HCC tissues and normal liver tissues,and whether they were significantly correlated with the overall survival and disease-free survival of HCC patients. The hub genes were conclusively identified,and their protein expression was validated through the Human Protein Atlas database. Results The genetic expression data in this study were obtained from 50 normal liver tissue samples and 373 HCC tissue samples. Through differential gene expression analysis,a total of 7 230 genes differential expression between HCC and normal hepatic tissue,comprising 3 691 up-regulated genes and 3 539 down-regulated genes in HCC were identified. Enrichment analysis showed that the up-regulated differentially expressed genes were mainly involved in cell cycle regulation and mitotic processes;the down-regulated differentially expressed genes were mainly involved in processes such as small molecule metabolism and organic acid metabolism. WGCNA identified 19 gene modules related to the clinical features of HCC patients,the cyan and purple modules were screened by analyzing the relationship between the modules and the clinical features. The first two genes in the cyan module genes that were strongly associated with both overall survival and disease-free survival of patients were VPS45 and FAM189B. In the purple module genes,first two genes that were strongly associated with both overall survival and disease-free survival of patients were CLEC1B and FCN3,respectively;therefore,VPS45,FAM189B,CLEC1B and FCN3 were identified as the final Hub genes. Immunohistochemical staining in the Human Protein Atlas database showed that VPS45 and FAM189B were expressed higher in HCC tissues than in normal liver tissues. FCN3 was expressed in HCC tissues lower than in normal liver tissues,the difference in the expression of CLEC1B between HCC tissues and normal liver tissues was not obvious. Conclusion VPS45,FAM189B,CLEC1B and FCN3 have been preliminary identified as possible novel potential biomarkers for HCC,which may provide a theoretical basis for targeted therapy of HCC.

  • Present and Future of Global Information Literacy: In Review of the European Conference on Information Literacy

    Abstract: [Purpose/significance] The paper reviews the European Conference on Information Literacy (ECIL) in the past five years, providing references for domestic scholars to understand the present situation and future trends of global information literacy. [Method/process] According to the author's experience of attending ECIL and conference materials & papers, the paper summarizes the conferences from the five aspects:the conference overview, the distribution of participating countries, the topics of the conference and the research hotspot, the various conference activities and the participating of Chinese scholars, to help domestic scholars understand the latest progress and the future development for information literacy. [Result/conclusion] The ECIL platform reflects the trend of global information literacy to some extent:focusing on the practical application,expanding the research field of information literacy education,extending more related concepts, enriching more target users and innovating new teaching process and means. The enthusiasm of Chinese scholars to participate in ECIL has been continuously improved. From the first two sessions, only scholars from Taiwan participated, but in 2017, scholars and postgraduates from mainland China participated in the ECIL in various ways. Scholars in related fields in the world have fully demonstrated the latest achievements and contributions made by Chinese universities and research institutes in the field of information literacy. Finally, it is recommended that the Chinese information literacy community draw on the advanced experience of ECIL in both content and form.

  • The Practice of Flipping Classroom Teaching of Science and Technology Information Retrieval Course Based on Rain Classroom

    Abstract: [Purpose/significance] Educational mode is facing a great change in the deep integration of learning technology and teaching. Actively adopting advanced teaching technology is the only way to reform and upgrade traditional curriculum. This paper summarizes the teaching practice of reversing the information retrieval course based on Rain Classroom, aiming at providing reference for other information literacy courses.[Method/process] Empirical research method is used to introduce the curriculum design from the aspects of teaching technology selection, teaching target orientation, learning time design, learning resources design, exercises design, classroom activities design, etc. The effect is evaluated by questionnaire survey.[Result/conclusion] The curriculum strengthens the practical application objectives, mobilizes students'enthusiasm for participation, strengthens the interaction between teacher and students, enriches the learning channels, cultivates students' autonomous learning ability and teamwork ability, and achieves good effectiveness.

  • Leveraging Advantages of University to Eradicate Poverty——Practice and Discussion on Targeted Poverty Alleviation Implemented by University of Science and Technology of China in Liuzhi Special District, Guizhou Province, China

    Abstract: Targeted poverty alleviation involved by universities is an important part of the poverty alleviation strategy in China, which reflects the responsibility of universities to serve the national strategy. This study has reviewed the specific practices, experiences and some achievements of poverty alleviation works conducted by University of Science and Technology of China (USTC) in Liuzhi Special District, Guizhou Province, China. According to actual situation of Liuzhi, USTC gives full play to the advantages of education and research, and carries out a series of poverty alleviation assistances by supporting party building, education, industrial development and sales of agricultural products. Over the past five years, USTC summed up its experience continuously and conducted a number of distinctive projects, which help Liuzhi achieve the goal of poverty alleviation and lay a solid foundation for rural revitalization.

    Abstract:本试验旨在研究条纹锯鮨(Centropristis striata)幼鱼维生素C需要量。试验采用单因素试验设计法,选取平均体重为(88.34±24.1) g的条纹锯鮨幼鱼630尾,随机分为7组,每组3个重复,每个重复30尾鱼。各组分别投喂维生素C含量(实测值)分别为10.97(对照)、74.03、148.33、213.64、335.33、618.88和910.98 mg/kg等氮等能饲料。检测饲料维生素C含量对条纹锯鮨幼鱼生长性能、血清生化指标、相关酶活性、各组织中维生素C积累量和肌肉成分的影响。试验期8周。结果表明:1)74.03 mg/kg组的增重率和特定生长率显著高于10.97、618.88和910.98 mg/kg组(P<0.05),74.03 mg/kg组的饲料系数显著低于10.97和910.98 mg/kg组(P<0.05),10.97和910.98 mg/kg组蛋白质效率显著低于74.03和618.88 mg/kg组(P<0.05),335.33 mg/kg组的肥满度显著低于74.03、148.33和213.64 mg/kg组(P<0.05)。2)10.97、74.03 mg/kg组血清总蛋白含量显著低于其他各组(P<0.05),618.88和910.98 mg/kg组血清甘油三酯含量显著低于其他各组(P<0.05),10.97 mg/kg组血清溶菌酶活性显著低于213.64、335.33、618.88、910.98 mg/kg组(P<0.05)。3)335.33 mg/kg组血清超氧化物歧化酶(SOD)活性显著高于10.97、74.03、910.98 mg/kg组(P<0.05),335.33 mg/kg组血清总抗氧化能力(T-AOC)显著高于10.97、618.88、910.98 mg/kg组(P<0.05),335.33 mg/kg组血清丙二醛(MDA)含量显著低于10.97 mg/kg组(P<0.05)。4)10.97 mg/kg组肝脏SOD和过氧化物酶(CAT)活性显著低于其他各组(P<0.05),148.33、335.33和910.98 mg/kg组肝脏MDA含量显著低于10.97 mg/kg(P<0.05)。5)74.03 mg/kg组肌肉SOD活性和T-AOC均显著高于其他各组(P<0.05)。6)213.64、335.33、618.88、910.98 mg/kg组血清谷丙转氨酶(GOT)活性显著低于10.97 mg/kg组(P<0.05),10.97 mg/kg组肝脏GOT活性显著低于其他各组(P<0.05)。10.97、74.03 mg/kg组血清谷草转氨酶(GPT)活性显著高于其他各组(P<0.05)。7)910.98 mg/kg组血清中维生素C积累量显著高于其他各组(P<0.05),910.98 mg/kg组肝脏中维生素C积累量显著高于10.97、74.03、148.33和213.64 mg/kg组(P<0.05),618.88 mg/kg组的肌肉中维生素C积累量显著高于10.97、74.03、148.33和213.64 mg/kg组(P<0.05)。8)74.03 mg/kg组肌肉水分含量显著低于10.97、148.33和213.64 mg/kg组(P<0.05),910.98 mg/kg组肌肉胶原蛋白含量显著高于其他各组(P<0.05)。由此可见,以特定生长率、肝脏中维生素C积累量和血清溶菌酶活性为评价标准,通过折线回归模型得出条纹锯鮨幼鱼饲料维生素C需要量分别为66.66、309.93和345.11 mg/kg

    Abstract:本试验旨在研究饲料中添加硒代蛋氨酸(Se-Met)对条纹锯鮨生长性能、组织硒含量、抗氧化能力及血清生化指标的影响。选取初始体重为(81.62±0.75) g的健康条纹锯鮨幼鱼540尾,随机分为6组,每组3个重复,每个重复30尾。各组分别投喂硒含量为0.09、0.21、0.44、0.58、0.77和1.05 mg/kg的试验饲料。试验期8周。结果表明:1)饲料硒含量为0.58 mg/kg组的终末体重和增重率最高,显著高于其他各组(P<0.05)。饲料硒含量为0.58 mg/kg组的饲料系数最低,显著低于饲料硒含量为0.09、0.77和1.05 mg/kg组(P<0.05)。2)饲料硒含量为0.77 mg/kg组的肌肉粗脂肪含量最高,显著高于除饲料硒含量为0.58 mg/kg组的其他各组(P<0.05)。饲料硒含量为0.44 mg/kg组的肌肉粗灰分含量最高,显著高于其他各组(P<0.05)。3)饲料硒含量为0.77 mg/kg组的肝脏中硒含量显著高于饲料硒含量为0.09、0.21和0.44 mg/kg组(P<0.05)。饲料硒含量为0.58 mg/kg组的血清中硒含量最高,显著高于饲料硒含量为0.09、0.21、0.44和1.05 mg/kg组(P<0.05)。4)饲料硒含量为1.05 mg/kg组的血清谷胱甘肽过氧化物酶(GPx)活性显著高于其他各组(P<0.05)。饲料硒含量为0.58 mg/kg组的血清谷胱甘肽还原酶(GR)活性最高,显著高于饲料硒含量为0.77和1.05 mg/kg组。饲料硒含量为0.58 mg/kg组的血清过氧化氢酶(CAT)活性最高,显著高于饲料硒含量为0.09、0.21和1.05 mg/kg组(P<0.05)。饲料硒含量为1.05 mg/kg组的血清谷胱甘肽S-转移酶(GST)活性最高,显著高于饲料硒含量为0.09、0.21、0.44和0.58 mg/kg组(P<0.05)。5)饲料硒含量为0.09 mg/kg组的肝脏GPx活性最低,显著低于饲料硒含量为0.44、0.58和0.77 mg/kg组(P<0.05)。饲料硒含量为0.09 mg/kg组的肝脏GR活性最低,显著低于饲料硒含量为0.44、0.58和1.05 mg/kg组(P<0.05)。饲料硒含量为0.09 mg/kg组的肝脏GST活性最高,显著高于饲料硒含量为0.44、0.58、0.77和1.05 mg/kg组(P<0.05)。饲料硒含量为0.77 mg/kg组的肝脏超氧化物歧化酶(SOD)活性最高,显著高于饲料硒含量为0.09和1.05 mg/kg组(P<0.05)。饲料硒含量为0.44 mg/kg组的肝脏CAT活性最高,显著高于其他各组(P<0.05)。6)饲料硒含量为0.58 mg/kg组的血清总蛋白含量最高,显著高于其他各组(P<0.05)。饲料硒含量为0.58 mg/kg组的血清总胆固醇含量最高,显著高于饲料硒含量为0.09、0.21和0.44 mg/kg组(P<0.05)。饲料硒含量为0.77 mg/kg组的血清免疫球蛋白M含量和溶菌酶活性最高,显著高于饲料硒含量为0.09、0.21、0.44和1.05 mg/kg组(P<0.05)。以增重率、肝脏中硒含量、血清溶菌酶活性为评价指标,通过二次曲线分析得出条纹锯鮨对硒的适宜需求量分别为0.62、0.92、0.72 mg/kg,可见饲料中硒的适宜含量为0.62~0.92 mg/kg。

  • SDN场景中基于双向流量特征的DDoS攻击检测方法

    Abstract: The distributed nature of traditional network resources makes it more difficult for administrators to realize the centralized control of the network. It is difficult to quickly and accurately detect and trace the DDoS attacks when distributed denial of service attacks occur. To solve this problem, combined with the advantages of centralized management and control of software defined network, the advantages of dynamic management and the characteristics of DDoS attacks, this paper first introduced the concept of bidirectional traffic feature, put forward the four-tuple characteristics of attack detection and made use of the growth hierarchical self-organizing map algorithm to analyze and classify the quaternion eigenvectors extracted from network flows quickly and accurately. At the same time, this paper proposed a new detection method that locates potential victims by adaptively changing the granularity of flow table. Simulation results show that the four-tuple features, as well as the detection algorithm issuing the monitoring flow entry, can detect DDoS attacks and pinpoint the victim with accuracy of nearly 96%, and the computational overhead for the controller is small.

  • 云纹石斑鱼幼鱼对高能低氮饲料的适应性

    Abstract:为研究云纹石斑鱼幼鱼对高能低氮饲料的适应性,采用双因素试验设计,设蛋白质水平分别为35%(P35)、40%(P40)、45%(P45),脂肪水平分别为9%(L9)、12%(L12)、15%(L15),共配制9种试验饲料,依次标记为P35L9、P35L12、P35L15、P40L9、P40L12、P40L15、P45L9、P45L12、P45L15。选取初始平均体重为27.09 g的云纹石斑鱼幼鱼810尾,随分为9组,每组设置3个重复,每个重复30尾,饲喂9周后测定其生长性能、肌肉营养组成和血清生化指标。结果表明:1)随着饲料蛋白质和脂肪水平的升高,云纹石斑鱼幼鱼的增重率和特定生长率先升高后降低,且饲料蛋白质和脂肪水平间有显著的交互作用(P0.05)。2)饲料脂肪水平对肌肉粗脂肪含量的影响显著(P0.05)。3)血清中谷草转氨酶活性随饲料脂肪水平的升高而升高,L15组活性最高(121.98 U/L),与L9组存在显著差异(P0.05);血清中尿素氮和饲料蛋白质水平呈显著的负相关(P0.05)。结果显示:在本试验条件下,饲料蛋白质水平为40%和脂肪水平为12%时云纹石斑鱼幼鱼的生长效果最好,饲料的利用率较高。

  • 饲粮营养水平对黄羽肉鸡生长性能、肉品质和性成熟的影响

    Abstract:本试验旨在研究饲粮营养水平对黄羽肉鸡生长性能、肉品质和性成熟的影响。采用单因子完全随机设计,将432只1日龄商品代北京油鸡随机分配到高、中和低3种营养水平组,每组4个重复,每个重复36只鸡,自由采食、饮水。饲粮按中速黄羽肉鸡的饲养标准分为3个阶段(1~5周、6~9周和10~13周),每个阶段在保持蛋白质与能量比值不变的情况下分别设置高、中和低3种营养水平。结果表明,1~5周,低营养水平组肉鸡的平均日采食量和平均日增重极显著低于中、高营养水平组(P0.05)。高营养水平组母鸡鸡冠重和鸡冠长度极显著高于中、低营养水平组(P0.05)。由此可见,对于中速型黄羽肉鸡,中营养水平的饲粮能获得较好的生长性能,高营养水平的饲粮能促进母鸡性成熟。
