  • Evaluation of long-term intervention effects of using smart sports bracelets in community-based type 2 diabetes patients

    Abstract: Background China accounts for 28% of the world's diabetic patients, but patient adherence to treatment is not high, which is related to the low level of specialization in lifestyle behavioral therapy. Training and education of dietitians and exercise prescribers have only started in China in recent years, and even fewer people have mastered the theory and implementation of behavioral interventions, and the number of practicing professionals is quite limited.Objective To construct a community intervention model for type 2 diabetes patients using smart sports bracelets and evaluate the long-term management effect. MethodsEach administrative district in Guangzhou selected one primary health care institution to recruit diabetes as study subjects. Intervention group and control group were matched by age and sex. Exercise was prescribed in accordance with the exercise guidelines for diabetes.The bracelet intervention group used sports bracelet for continuous follow-up management, while the control group used conventional management mode. Both groups were followed up one year later. Results After 1 year, compared to baseline, HbA1c compliance rate increased by 12.74% (P=0.02) in bracelet intervention group. Both self-management and diet management behavior scores were significantly increased in intervention group, and waist circumference was significantly reduced (all P<0.05). Fasting blood glucose, triglyceride and low density lipoprotein cholesterol in control group were improved (all P<0.05). After 1 year, the HbA1c compliance rate was significantly higher in the intervention group than in the control group (P=0.03).After adjusting for age, sex and family history of diabetes, duration of disease, triacylglycerol and apolipoprotein B were found to be influential factors for elevated HbA1c (all P < 0.05).After adjusting for age, sex and family history of diabetes, duration of disease, triacylglycerol and apolipoprotein B were found to be influential factors for elevated HbA1c(all P<0.05).ConclusionUsing a smart sports bracelet helps people with diabetes maintain self-management behaviors over time and keep their blood sugar at target.Dyslipidemia is an important factor affecting blood glucose control and should be paid attention to. On the basis of lifestyle modification, pharmacological interventions should be initiated as early as possible to regulate lipid metabolism disorders.

    Insomnia is the most common concomitant symptom in patients with depressive disorder. Early and effective evaluation of insomnia symptoms in patients with depression is the key to prevent sleep problems and further deterioration of depression. This paper expounds the regional homogeneity of resting-state functional Magnetic Resonance Imaging used in patients with depressive disorder and insomnia disorder, and points out the difference between patients with unipolar depression disorder(UDD)and bipolar depression disorder(BDD), so as to provide reference for the clinical diagnosis, differentiation, diagnosis and treatment of patients with unipolar depression and bipolar depressive disorder Intervention and related research provide theoretical basis and practical reference.

    Abstract: " Prior research has consistently demonstrated that consumers have a negative preference for brands associated with dissociative groups. Conditions under which these preferences might be changed have remained largely unexamined. The current work investigates the effect of threat of freedom on consumers’ preferences for brands associated with dissociative groups. We propose that consumers who experience a high level of threat of freedom will prefer brands associated with dissociative groups. This effect is mediated by a heightened feeling of psychological reactance induced by the high level of threat of freedom. We also examined the moderating role of narrative and self-esteem on the effect. One pilot experiment and three laboratory experiments were conducted to test our conceptualization. In experiment 1, we assessed the basic effect by which threat of freedom influences consumers’ preferences for brands associated with dissociative groups. In this experiment, we also examined the mediating effect of psychological reactance. In experiment 2, we tested the robustness of the effect found in experiment 1 and investigated the moderating role of means of narrative through a two-by-two inter-subject design (freedom threat: low vs. high and means of narrative: yes vs. no). In experiment 3, we investigated the moderating role of self-esteem through a two-by-two mixed design (freedom threat: low vs. high and self-esteem: low vs. high). The results of three experiments confirm our predictions. The results of experiment 1 showed that the participants in the high threat-of-freedom group tended to prefer brands associated with dissociative groups. This effect was found to be mediated by psychological reactance. In experiment 2, our findings showed that the influence of threat of freedom on consumers’ preference for brands associated with dissociative groups was replicated when a non-narrative information transmission model was used, and the effect was mitigated when a narrative information transmission mode was used. Finally, experiment 3 confirmed the moderating role of self-esteem by showing that participants with high self-esteem tended to prefer brands associated with dissociative groups when they experienced high vs. low threat of freedom. For participants with low self-esteem, such effects became non-significantly different. The current research has theoretical and practical contributions in the following aspects. First, it contributes to the literature of dissociative groups by documenting a condition in which consumers’ negative attitude toward brands associated with dissociative groups can be improved. Second, this research extends the current literature on threat of freedom by revealing a preference for brands associated with dissociative groups as a new behavioral downstream. Finally, our findings have clear marketing implications. Certain types of consumers may experience varying levels of threat of freedom. Marketers may consider using actors who represent a dissociative identity in their marketing communications to attract this type of consumers.

  • 海藻多糖通过下调肝癌细胞Hep3B糖酵解途经抑制细胞增殖和迁移

    Abstract:摘要 目的:探讨海藻多糖(Algal Polysaccharides,AP)对肝癌细胞Hep3B的增殖和迁移的影响及其可能的作用机制,为治疗肝癌提供新的思路。方法:1)比较正常细胞与癌细胞中糖酵解途经(Embden-Meyerhof-Parnas,EMP)的表达差异,用紫外分光光度计比色法检测EMP限速酶酶活力:已糖激酶(Hexokinase, HK),丙酮酸激酶(Pyruvate kinase, PK), 用乳酸测定试剂盒测定EMP产物:乳酸;2)AP处理Hep3B后,检测EMP表达水平变化;3)AP处理细胞后,检测肝癌细胞Hep3B细细胞的增殖和迁移能力及上皮细胞-间充质转化(EMT)的影响,方法包括MTT、qPCR和Transwell小室实验;4)MTT、Transwell和qPCR检测HK抑制剂3-溴丙酮酸(3-Bromopyruvate,3-BrPA)对肝癌Hep3B细胞的活力和迁移能力及EMT的影响;5)Western Blot和相关试剂盒检测海藻多糖与3-BrPA分别处理细胞时,对EMP水平以及Akt通路的影响;6)AP联合3-BrPA处理HepB3后,检测HK和Akt信号通路表达水平及细胞活力和迁移的变化。结果:1) 癌细胞(Hep3B,Hela,SW480)EMP代谢水平均高于正常肝细胞(HL02),其中Hep3B差异最为显著;2) AP能抑制 Hep B细胞 EMP代谢水平,且抑制程度随着AP浓度升高而升高,存在浓度依赖性;3) AP可抑制肝癌 Hep3 B细胞的增殖和迁移,且抑制EMT的发生。AP浓度为200 mg/ mL,处理时间为48 h时,生长抑制率达(55±2.8)%,细胞迁移数为对照组的(32±2.9)%;4) 3-BrPA可下调Hep3B细胞HK活性,并抑制细胞活力与迁移,且抑制EMT的发生。200 μmol/ L3- BrPA作用细胞48 h后,与对照组相比,细胞活性下降了(52±5.8)%( p<0.001),细胞迁移数下降了(48±6.1)%( p<[0.01]); 5) 海藻多糖和3-BrPA均下调糖酵解代谢水平,且抑制Akt信号通路; 6) AP与3-BrPA联合处理Hep3B后,HK表达下降,显著抑制Akt信号通路,细胞活性和迁移能力均较海藻糖单用组显著下降(p<0.05)。结论:肝癌 Hep3 B细胞 EMP代谢水平高于正常肝细胞,海藻多糖可下调 Hep3B细胞 EMP代谢水平,低EMP代谢水平可能通过下调EMT和Akt信号通路抑制Hep3B的增殖和迁移。当AP和3- BrPA联合应用时,抑制肝癌迁移和增殖效果更明显。

  • 卤地菊乙醇提取物W40单体诱导GLC-82细胞凋亡的分子机制研究

    Abstract:目的:探讨卤地菊Wedelia prostrate(Hook.et Arn.)Hemsl.的乙醇提取物W40单体对非小细胞肺癌GLC-82细胞的抗肿瘤作用分子机制。方法: 通过MTT和克隆形成抑制实验检测W40对非小细胞肺癌GLC-82细胞增殖的影响; 通过细胞划痕实验检测W40对细胞迁移的影响; 通过Annexin V-FITC /PI 双染法检测W40对细胞凋亡的诱导; 通过Western blotting分析细胞增殖和凋亡相关蛋白的水平。结果: MTT实验表明,W40对GLC-82细胞有较为明显的细胞毒作用;克隆形成抑制实验表明,W40可以显著抑制GLC-82细胞增殖, 且抑制程度呈现浓度依赖效应;细胞划痕实验表明,W40能够一定程度上抑制GLC-82细胞的迁移能力; Annexin V-FITC /PI 双染的结果显示,随着药物浓度的增加, 细胞凋亡率逐渐增加; Western blotting 结果表明,随着药物浓度的增加, p-Stat3蛋白的表达水平下降, Stat3下游蛋白Bcl-2、Mcl-1的表达减少。同时,促凋亡蛋白Bax的表达增多, 并伴随Caspase-9、Caspase-3 活化及多聚ADP- 核糖聚合酶PARP 酶切失活;p-BRAF蛋白的表达水平下降, p-BRAF下游蛋白p-MEK、p-ERK表达减少。结论: W40 通过抑制BRAF / MAPK / ERK 及Stat3信号通路来诱导细胞凋亡。
